Abstract

Fast dissolving tablets of Metformin hydrochloride were prepared by direct compression method. FDTs of Metformin hydrochloride were formulated by using super disintegrate like Croscarmellose sodium (4%) and sodium starch glycolate (8%). Hardness for the formulations F1 to F6 was found to be in the range of 4.5±0.08 to 3.7±0.10 Kg/cm2. Disintegration time was found to be in the range 61.5±0.54 to 24.5±1.04 sec. Formulations passed the friability and weight variation tests. It was also found that with the increase in concentration of super disintegrates the hardness and disintegration decreased. Out of all six formulations F6 showed the best features with least disintegration time of 24.5±1.04 seconds. It was due to effect of natural super disintegrates and the mechanism involved. The in-vitro release was studied for all the formulations and the drug release ranged from 97.40±2.46 to 99.54±1.12 respectively. The evaluation parameters showed that the use of natural super disintegrates showed better hardness and disintegration time, wetting time and drug release as compared to synthetic super disintegrates. From the above results, it is concluded that formulations F6 shows better results with proper hardness of 3.7±0.10 kg/cm2, disintegration time of 24.5±1.04 seconds, 0.44±0.01% friability, wetting time 17.1±0.76 sec, dispersion time 16.3±0.8 sec, drug content 100±1.2%. It fulfils all the requirements for fast dissolving tablets. This formulation showed maximum drug release of 99.54±1.12. Formulation F6 was found to better than other formulations due to less disintegration time and wetting time with proper hardness as compared to other formulations consisting of synthetic and natural super disintegrates.
