Simethicone, an anti-flatulent medication, is often formulated as a suspension for ease of administration. However, its limited solubility and slow dissolution rate can hinder its therapeutic efficacy. In this study, we aimed to develop and evaluate a simethicone nanosuspension to overcome these challenges and enhance its bioavailability. The nanosuspension was prepared using the precipitation method, with polyvinylpyrrolidone K30 (PVP K30) and sodium lauryl sulfate (SLS) identified as suitable stabilizers through compatibility studies. Various ratios of drug to stabilizer and drug to polymer were investigated, with the optimized batch (NS5) achieving a mean particle size of 258 nm and a zeta potential of -21.8 mV. Stability studies confirmed the formulation's robustness under different conditions. In vitro dissolution studies revealed a remarkable 84 percent increase in drug release within 12 hours compared to a coarse suspension. Particle size reduction was identified as the primary factor contributing to this accelerated dissolution rate. These findings suggest that nano-precipitated simethicone nanosuspensions may offer superior therapeutic benefits over conventional formulations, highlighting their potential for enhanced bioavailability and improved patient outcomes.
