Abstract: The objective of the present work is to prepare spherical polyelectrolyte microcapsules of Zidovudine by a means of complex coacervation method in an emulsion system. Chitosan was used as the cationic polymer and sodium alginate and acacia as the anionic polymers. The two polymers were made to interact at a concentration of 2% and the coacervation was achieved at pH6.8. Glutaraldehyde was used as the cross linking agent. Microspheres were prepared in drug to polymer ratios of 0.5:1, 0.75:1 and 1:1. Microsphere suspension in arachis oil was prepared in ratio of 0.5:1. The emulsification used for microparticle formation allows the preparation of particles with spherical shape and narrow size distribution. The suspension was better than the dry product with respect to shape, drug content and also order of release, which was zero order for suspension whereas first order for the dried product. The drug from the suspension formulation was found to release over a period of 10hrs. The oily suspension provides a means of parenteral administration of Zidovudine, provided the particle size can be further reduced. This sustained release parenteral formulation of Zidovudine would be beneficial in comparison with the current intermittent oral dosage regimen.
