Gestational Diabetes Mellitus (GDM) is one of the major global public health issues in Asia. Various genetic and environmental factors are found to be accountable for the development of GDM. In this back ground SLC30A8 rs13266634 polymorphism is hypothesized to increase the risk of GDM in South-Indian population. The present study aimed to evaluate the association of SLC30A8 rs13266634 polymorphism with risk of GDM. A total of 400 participants, 200 GDM and 200 healthy pregnant women were included in the case-control study. Genotyping was performed by polymerase chain reaction–restriction fragment length polymorphism (PCR-RFLP). The genotype and allele frequencies were analyzed by applying appropriate statistical methods. An increased frequency of TT genotype and T allele of SLC30A8 rs13266634 polymorphism was observed in GDM women. There was no significant difference in the insulin c-peptide levels between GDM and controls (p=0.2). The minor allele 'T' displayed 1.71 fold (OR=1.71; CI 1.24-2.36, p=0.001) increased risk of GDM. Our study revealed a significant association of SLC30A8 rs13266634 polymorphism with GDM in South-Indian population.
